ABSTRACT

Objective To explore the epidemiological characteristics of pulmonary infection in elderly patients with chronic obstructive pulmonary disease (COPD), and to construct a risk prediction model. Methods Among of 125 elderly patients with COPD from May 2020 to June 2022 were selected as the research subjects. The epidemiological characteristics of infected patients were counted, and the risk factors of pulmonary infection in patients were analyzed and a prediction model was constructed. Results A total of the 125 elderly patients with COPD, there were 46 cases of pulmonary infection, with the infection rate of 36.80%. The detection rate of Gram-negative bacteria was higher than that of Gram-positive bacteria or fungi (64.44% vs 33.33% or 2.22%, P2=0.812 and P=0.295. ROC curve analysis revealed that the AUC value of the prediction model on predicting the pulmonary infection in elderly patients with COPD was 0.802. Conclusion The pathogenic bacteria of elderly patients with COPD complicated with pulmonary infection are mainly Gram-negative bacteria. The prediction model constructed according to the risk factors of pulmonary infection in patients has predictive value on pulmonary infection in patients.

ABSTRACT

Objective To analyze Pseudomonas aeruginosa (PA) infection and drug resistance in the elderly with respiratory tract infection, so as to provide a basis for the control of nosocomial infection and rational use of antibiotics. Methods The samples from elderly inpatients with respiratory tract infection were collected between March 2020 and March 2022. PA infection/colonization were investigated, and the drug resistance of pathogens was determined according to CLSI criteria (2019 version). Results There were 123 strains of PA isolated from the sputum and bronchoscopy lavage fluid of elderly patients with respiratory tract infection. The main departments with positive PA detection were respiratory department, ICU ward and neurology department. The difference of PA detection in different years was not statistically significant (P>0.05). The proportion of nosocomial infection in 2021 was lower than that in 2020 (44.44% vs 63.33%, c2=4.410, P=0.036). The resistance rate of 123 isolated PA strains to piperacillin was >90.00%, and they were resistant to ceftazidime, cefotaxime, cefepime, aztreonam and gentamicin to varying degrees. There was no significant difference in resistance rate of PA to antibiotics in different years (P>0.05). In the 123 strains of pathogens, there were 17 strains (13.82%) of carbapenem-resistant PA, and their resistance to common antibiotics was significantly higher than that of carbapenem-sensitive PA (P<0.05). Conclusion The main pathogen of nosocomial infection is PA, and the proportion of nosocomial infection shows a downward trend. The detection rate of carbapenem-resistant PA is high. In clinical treatment, targeted antibiotics can be applied.

ABSTRACT

OBJECTIVE@#To analyze the clinical phenotype and genetic basis for a Chinese pedigree suspected for branchiootic syndrome (BOS).@*METHODS@#The proband was subjected to target-capture high-throughput sequencing to detect potential variant of deafness-associated genes. Candidate variants were verified by Sanger sequencing of the family members.@*RESULTS@#The proband was found to harbor a c.1627C>T (p.Gln543Ter) nonsense variant of the EYA1 gene. Sanger sequencing confirmed that all of the 4 patients with the BOS phenotype from the pedigree have harbored the same heterozygous variant. Based on the guidelines of the American College of Medical Genetics and Genomics, the variant was predicted to be pathogenic (PVS1+PS+PP3+PP4).@*CONCLUSION@#The c.1627C>T (p.Gln543Ter) variant of the EYA1 gene probably underlay the BOS phenotype in this pedigree. Above finding has provided a basis for its clinical diagnosis.

ABSTRACT

Objective:To improve the diagnostic criteria of suspected cases through investigating the epidemiological and clinical manifestations of confirmed cases of novel coronavirus infection in children.Methods:We retrospectively analyzed the epidemiological and clinical manifestations of six children with novel coronavirus infection diagnosed in Chongqing Three Gorges Central Hospital from February 3, 2020 to February 15, 2020.Compared with the diagnostic criteria of suspected cases, we summarized the problems encountering in the application of this standard in clinic and try to put forward suggestions for improvement.Results:Among the six confirmed cases: five males and one female; three from Hubei Province and three from Wanzhou; six cases of clustered onset of the family.Visiting nature: six cases of suspected case, six cases from community or outpatient screening.Three fever cases with/without respiratory symptoms, one of which had diarrhea; all children′s blood routine and lymphocyte counts were within the normal range; chest CT imagings of No.1 and No.5 case showed typical novel coronavirus pneumonia signs, and the other three patients had abnormalities without the characteristics of novel coronavirus pneumonia, and one case was normal.Comparison results: Only No.1 case fully met the diagnostic criteria, and the remaining cases did not meet the diagnostic criteria for early suspected cases.Conclusion:In order to improve the accuracy and practicality of the diagnosis of suspected cases in children, it is recommended to refine and standardize the diagnostic criteria for some suspected cases.

ABSTRACT

Objective@#To understand status of amphetaminetype stimulants (ATS) use among residents aged 15-64 in a border city of Yunnan province.@*Methods@#Using the stratified cluster random sampling method, a total of 3 130 residents were recruited through both anonymous questionnaire and interview, regarding their health-related behaviour and ATS use.@*Results@#Among 3 130 residents aged 15-64 years in this city, the overall prevalence rates of ATS use were 4.0% (126/3 130) in the lifetime and 2.6% (82/3 130) in the past one year, while the prevalence of ATS use disorder in the past year was 2.3% (73/3 130). The prevalence rates of lifetime ATS use and in the past year were 7.5% (108/1 443) and 5.1% (73/1 443) in the high epidemic area, 7.3% (122/1 682) and 4.8%(80/1 682) in males, 5.2% (118/2 260) and 3.4% (77/2 260) in 18-45 age group, 4.6%(63/1 361) and 3.4% (46/1 361) in the ones having had elementary school education, 10.3% (50/487) and 8.6% (42/487) in unmarried group, 17.1% (19/111) and 12.6% (14/111) in either divorced, widowed or separated group, 4.8% (108/2 256) and 3.2% (72/2 256) in the farmers group, 6.0% (99/1 643) and 4.4% (73/1 643) in the non-religious groups (neither Buddhism nor Christianity), 15.2% (97/639) and 11.9% (76/639) in groups with negative hobbies which were 1.3% (29/2 314) and 0.3%(6/2 314) higher than the ones with positive hobbies, 8.5% (84/992) and 6.3% (62/992) in the ones negatively managing the stress which were 2.0% (42/2 138) and 0.9% (20/2 138) higher than the ones that treating the stress in a positive way.@*Conclusions@#The prevalence rates of ATS use and ATS use disorder in the past one year appeared high among those residents aged 15-64 years in the border city of Yunnan province. Prevention and intervention programs should be carried out for the target groups as soon as possible.

ABSTRACT

Objective To investigate the expression of LRG1 (Leucine-rich alpha-2-glycoprotein 1) and CD105 (Endoglin) in normal cervical tissues , cervical intraepithelial neoplasia (CIN)Ⅱ- Ⅲ and cervical carcinoma. Furthermore, to explore the function of LRG1 in cervical carcinoma pathogenesis and the relationship between LRG1 and microvessel density.Methods The expression levels of LRG1 and CD105 were detected by immunohistochemistry in 40 cervical carcinoma tissues, 20 CINⅡ- Ⅲ tissues and 20 normal cervical tissues. Results Compared to the normal cervix, the expression of LRG1 in CINⅡ- Ⅲ and cervical squamous cell carcinoma was significantly increased (P<0.05).LRG1expression was correlated to clinical stage and lymph node metastasis (P<0.05) . But there was no correlation between LRG1 expression and age, the size of tumor, pathologic grades and depth of invasion (P>0.05).With the deepening of cervical lesions, CD105-MVD (X± s) increasedsignificantly (P<0.05).The expression of CD105-MVD in cervical carcinomawas related to clinical stage, lymph node metastasis and the size of tumor. The difference was statistically significant (P<0.05) . There was no correlation between CD105-MVD and age, pathologic grades and depth of invasion (P>0.05).Positive correlation was found between the expression of LRG1 and CD105-MVD (r=0.944,P<0.05).Conclusions The expression of LRG1 and CD105-MVD is up-regulated and shows a positive correlation, which suggests that the abnormal expression of LRG1 and CD105-MVD may involve in the occurrence and development of cervical squamous carcinoma.

ABSTRACT

<p><b>OBJECTIVE</b>To explore the genetic cause for a Uyghur Chinese child with collodion skin.</p><p><b>METHODS</b>G-banded chromosomal karyotyping was carried out for the child and his parents. High-throughput sequencing for 25 genes related to ichthyosis and ichthyosiform dermatosis was also performed for the child.</p><p><b>RESULTS</b>No karyotypic abnormality was found in the child and his parents. High-throughput sequencing has detected in the patient a previously described pathogenic mutation c.919C>T (p.Arg307Trp) and a novel c.856C>T (p.Arg286Trp) mutation in the TGM1 gene. By Sanger sequencing, the child was verified to have carried both mutations. His father was found to be a heterozygous carrier of the c.856C>T (p.Arg286Trp) mutation, while neither mutation was found in the mother.</p><p><b>CONCLUSION</b>Congenital ichthyosis associated with the TGM1 gene may show an autosomal recessive inheritance. The collodion condition of the child is probably due to the compound heterozygous mutations of the TGM1 gene.</p>

ABSTRACT

Epilepsy is a chronic neurological disorder with different etiologies. Despite of many antiepileptic drugs (AEDs) used in clinical treatment, about 30% of the patients have uncontrolled onset, and combined use of different AEDs or new AEDs are needed. The new AEDs should have satisfactory clinical efficacy, tolerability and pharmacokinetic properties, and minimal adverse effects. By now there have been about 20 novel AEDs; most of them are derivatives or analogues of current AEDs. Brivaracetam (BRI) is the derivative of levetiracetam (LEV). This review summarizes the mechanisms of action, pharmacokinetic profiles, tolerability, pharmacology and toxicology, and side effects of LEV and its derivative BRI.

ABSTRACT

<p><b>OBJECTIVE</b>The aim of this study was to transfect TPT1 into cell lines SMMC-7721 and L-02, seperately, and to observe the changes of biological behaviors of the cell lines.</p><p><b>METHODS</b>Through lipofectamine, the eukaryotic report expression vector containing TPT1 ORF (open reading frame), pEGFP-N3TPT1, were transducted into hepatocarcinoma cell line SMMC-7721 cells and normal liver cell line L-02 cells, seperately. The transduction was repeated three times in 24 hrs. The differences of biological behaviors between the pEGFP-N3TPT1 and pEGFP-N3 groups were studied by RT-PCR, MTT assay, soft agar colony formation assay and cell cycle analysis.</p><p><b>RESULTS</b>The pEGFP-N3TPT1 transfected cells had a high mRNA level in the two cell lines (P < 0.05) compared with the pEGFP-N3 controls. The ability of proliferation and the soft agar colony formation were enhanced in the SMMC-7721 transducted cells with pEGFP-N3TPT1 compared with that transducted with pEGFP-N3 (P < 0.05), and the cell cycle analysis showed that the cells in the phase G(2)+S/M increased after pEGFP-N3TPT1 transduction. In the L-02 cell line, we obtained similar results, pEGFP-N3TPT1 enhanced the colony formation in plate (P < 0.05), but not make it form colony in soft agar.</p><p><b>CONCLUSIONS</b>TPT1 can enhance malignant phenotype of SMMC-7721 cells and promote the growth of L-02 cells, but not transform L-02 into malignant phenotype.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the effects of autophagy gene Beclin 1 on growth of cervical cancer HeLa cells in vitro and vivo.</p><p><b>METHODS</b>The eukaryotic expression vector of Beclin1 was constructed and transfected via lipofectamine into HeLa cells. The experimental cells were classified into 3 groups: pcDNA3.1(+)-Beclin1 group,pcDNA3.1(+) group and HeLa group. Real time-ploymerase chain reaction and Western blot were used for detecting expression of Beclin1 mRNA and protein in the transfected cells. Flow cytometry (FCM) was employed to observe the effect of transfection on the apoptosis of HeLa cells, and proliferation was analyzed by MTT assay. The formation of autophagic vacuoles was measured by MDC staining. HeLa cells transfected with plasmid pcDNA3.1(+)-Beclin1 and pcDNA3.1(+) were inoculated subcutaneously in nude mice. The carcinogenic and growth activities of cancer cells in vivo were observed.</p><p><b>RESULTS</b>Eukaryotic expression vector pcDNA3.1(+)-Beclin1 was constructed successfully. It significantly improved the expression of Beclin1 mRNA and protein in HeLa cells. The proliferation of HeLa cells was inhibited, and the inhibition rate was 58.7%. FCM investigation showed that the apoptotic rate was (28.22 ± 2.34)% of pcDNA3.1(+)-Beclin1 group, significantly higher than the (14.6 ± 4.6)% in the pcDNA3.1(+) group and (11.2 ± 3.0)% in the HeLa group (P < 0.05). The monodansylcadaverin (MDC) staining showed significantly more autophagic vacuoles in the pcDNA3.1(+)-Beclin1 group (10.9%) than that in the pcDNA3.1(+) group (3.1%) and HeLa group (2.5%) (P < 0.05). After transfected with vector pcDNA3.1(+)-Beclin1, the carcinogenic activity of HeLa cells was decreased in nude mice, and the inhibition rate of tumor growth was 52.2%.</p><p><b>CONCLUSIONS</b>Autophagy gene Beclin 1 overexpression can inhibit the proliferation and growth of HeLa cells in vitro and vivo,while promote autophagy and apoptosis of HeLa cells. So it might be one of new gene therapy strategies for cervical carcinoma.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To explore the effect of Beclin1 overexpression on the growth of ovarian carcinoma cell line SKOV3 in vitro and in vivo.</p><p><b>METHODS</b>The recombinant plasmid pcDNA3.1/Beclin1 was constructed and transfected into SKOV3 cells via lipofectamine 2000. MTT assay was used to evaluate the effect of Beclin1 overexpression on the proliferation and growth of the transfected cells, whose apoptosis and autophagy were analyzed by flow cytometry. SKOV3 cells transfected with the plasmids pcDNA3.1/Beclin1 or pcDNA3.1 were inoculated subcutaneously in nude mice, and their carcinogenic and growth activities in vivo were evaluated.</p><p><b>RESULTS</b>MTT assay showed that transfection with pcDNA3.1/Beclin1 significantly inhibited the proliferations of SKOV3 cells, with a cell inhibition rate of 58.68% (P<0.05). The transfection also resulted in a cell apoptosis rate of (21.26-/+3.89)%, significantly higher than that of pcDNA3.1 trasnfection (P<0.05). Flow cytomerty showed that pcDNA3.1/Beclin1 transfection of SKOV3 cells produced a significantly higher MDC fluorescent intensity than pcDNA3.1 transfection. The SKOV3 cells transfected with vector pcDNA3.1/Beclin1 also showed decreased carcinogenic activity in nude mice, with a growth inhibition rate of 50.27%.</p><p><b>CONCLUSION</b>Beclin1 overexpression can inhibit the proliferation and growth of SKOV3 cells in vitro and vivo, suggesting its potential role in gene therapy of ovarian carcinoma.</p>

ABSTRACT

An improved everted gut sac method was applied to the study of prescription compatibility effect on the major components in Danshen extracts. With the separation and detection by HPLC-ECD, 5 major peaks could be detected in intestinal absorbed solution after prescription administration. Following the identification by HPLC-MS/MS, peak 2, 3, 4, and 5 were rosmaric acid, lithospermic acid, salvianolic acid B, and salvianolic acid A, respectively, which also confirmed with reference standards of those components. Through paralleling substance identification, peak 2, 3, 4, and 5 could be found as the major components in Danshen extracts, except Salvianolic acid E which is undetectable in intestinal solution. The contents of peak 2, 3, and 4 did not show difference before and after compatible prescription administrated, where the peak 5 had a significant increase in the same process. Those results revealed that peak 5, salvianolic acid A, might lead to an increasing pharmacological effect after prescription compatibility.

<p><b>OBJECTIVE</b>The aim of this study was to determine the metameric color differences between natural teeth and three brands of commercially available resin teeth.</p><p><b>METHODS</b>The spectral reflectance and color coordinates of natural teeth and three brands of commercially available resin teeth of A2 shade were measured with a spectrophotometer (PR-650) according to the CIE L*, a*, b* and CIE XYZ color scale relative to illuminant D65, A, cool white fluorescent (CWF) and ultraviolet (UV), and the metameric indices were calculated to determine the metameric color differences between natural teeth and resin teeth.</p><p><b>RESULTS</b>CIE L*, a*, b* values were influenced by the type of illuminants in both natural teeth and resin teeth. The pattern of spectral reflectance curves for natural teeth and resin teeth of A2 shade were different, while there were more than three crossing points among each curves, which meant the color of natural teeth and resin teeth of A2 shade might be the same under certain illuminant. The metameric indices between natural teeth and resin teeth of A2 shade were 3.48, 2.52 and 3.36 under illuminant A; 1.21, 1.90, and 2.79 under illuminant CWF; 1.59, 2.07, and 4.07 under illuminant UV. The metameric indices between resin teeth of different brand were 1.08, 0.10, and 1.01 under illuminant A; 1.46, 2.23, and 0.94 under illuminant CWF; and 2.55, 2.69, and 4.64 under UV.</p><p><b>CONCLUSION</b>Changes in optical properties of resin teeth of A2 shade relative to the different illuminants were different from those of natural teeth, the metameric effect between natural teeth and resin teeth of A2 shade were significant. Therefore, shade matching between natural teeth and resin teeth should be performed under more than one illuminant.</p>

ABSTRACT

A sensitive liquid chromatography-tandem mass spectrometric (LC-MS/MS) method was applied for the quantification of each component: tetrahydropalmatine (THP), dehydrocorydaline (DHC), salvianolic acid B (SAB), ginsenoside Rg1, Re, Rb1 and Rd in the Chinese herbal component SSTG (Shuangshentongguan) with different combinations. The pharmacokinetic data were analyzed with WinNonlin 5.2 software. The results showed that combination can increase the THP AUC value while the AUC values of SAB, ginsenoside Rg1, Re, Rb1 and Rd were reduced. These results showed significant differences. The AUC value of ginsenoside Rb1 was increased when combined with Danshen or Yanhusuo, but reduced when combined with Danshen and Yanhusuo. The DHC concentration in serum was too low to be determined.

ABSTRACT

<p><b>OBJECTIVE</b>To detect the expession of THY1 in ovarian serous cystadenocarcinoma tissues.</p><p><b>METHODS</b>Immunohistochemistry was performed to detect the expression of THY1 gene in formalin-fixed, paraffin-embedded specimens of normal ovaries (n = 25), ovarian serous cystadenoma (n = 25), and serous cystadenocarcinoma (n = 53). The correlation of THY1 expression with clinicopathological parameters was statistically analyzed.</p><p><b>RESULTS</b>The positive expression rates of THY1 protein in normal ovaries, ovarian serous cystadenomas and ovarian serous cystadenocarcinomas were 60.0% (15/25), 72.0% (18/25) and 34.0% (18/53), respectively. The values of IOD of THY1 protein expression were 288,449.2 +/- 60,087.3, 271,655.6 +/- 66,588.7 and 252,087.6 +/- 45,559.4, respectively. The expression of THY1 protein was significantly down-regulated in ovarian serous cystadenocarcinoma tissues compared with that in normal ovarian tissues and ovarian serous cystadenoma tissues (P < 0.05). THY1 expression was negatively correlated with surgical-pathological staging, histological differentiation and lymph node involvement (P < 0.05).</p><p><b>CONCLUSION</b>The decreased level of THY1 expression may be related with the occurrence and development of ovarian serous cystadenocarcinoma.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the bond strength of total-etch or self-etch dentin bonding agents after using two different dentin desensitizers on exposed dentin and investigate the bond interface by scanning electron microscope (SEM).</p><p><b>METHODS</b>Thirty intact and non-carious human third molars were used. The occlusal enamel was removed with the use of a slow-speed saw under water cooling. These teeth were divided into three groups using a table of random numbers with 10 teeth each. These three groups were treated with water (Group C), UltraEZ (Group U) and MI Paste (Group M) respectively. Then 10 teeth from each group were divided into A subgroup (n = 5) bonded with Single Bond 2 adhesive system and B subgroup (n = 5) bonded with Xeno III adhesive system according to manufacturers' instructions. A block of composite resin was build up to 4-5 mm. All the teeth were sectioned occluso-gingivally to obtain bar-shaped specimens with bonded surface area about 0.9 mm x 0.9 mm. The tension of the sample was tested by a microtensile tester at 1 mm/min. The mean values of bond strength were compared using one-way ANOVA. Three samples were chosen randomly from each of six groups for SEM investigation.</p><p><b>RESULTS</b>There were no significant differences between Group U and Group C both in A and B subgroups. While there were significant differences between Group M and Group C in two bonding-agent subgroups. For SEM, the hybrid layer was thin and dense in six groups. Both total-etch and self-etch bonding systems could get fair resin tag infiltration in Group C and Group U. In Group M, the resin tags were relatively shorter and fewer than the anterior mentioned two groups.</p><p><b>CONCLUSIONS</b>UltraEZ had no effect on bond strength of both kinds of dentin bonding agents, while MI paste could diminish bond strength.</p>

ABSTRACT

A sensitive liquid chromatography-tandem mass spectrometric (LC-MS/MS) method was developed and validated for the quantification of tetrahydropalmatine (THP) and dehydrocorydaline (DHC) in rat plasma. The compounds were simply pretreated by protein precipitation using acetone. Chromatographic separation was achieved on a reversed-phase SB-C18 column with the mobile phase of acetonitrile-ammonium acetate (0. 1% acetic acid) and step gradient elution resulted at a flow rate of 0.80 mL x min(-1). A tandem mass spectrometer equipped with electrospray ionization source was used as detector and operated in the positive ion mode. Quantification was performed using multiple reaction monitoring (MRM) of the transitions m/z 356. 2 --> m/z 191.9 and m/z 366. 2 --> m/z 350.2 for THP and DHC respectively. The method showed excellent linearity over the concentration range 1-1 000 ng x mL(-1) of two components (r = 0.994 for THP and r = 0.992 for DHC). The low limits of quantification were both 1 ng x mL(-1). The extract recoveries of analytes were from 71.71% to 91.59% for THP and from 83.27% to 103.15% for DHC. The precisions, the accuracy and the stability of the analytes meet the requirements. The method was applied to a pharmacokinetic study of THP and DHC after oral administration of the total alkaloid extraction of Rhizoma Corydalis (Yanhusuo). The AUC were (1.90 +/- 0.04), (2.58 +/- 0.08) and (4.34 +/- 0.19) mg x L(-1) h for low, medium and high doses of THP, respectively. While the DHC concentrations in plasma of low dose and medium dose were too lower to be detected, the AUC of high dose was (0.0896 +/- 0.0002) mg x L(-1) h.
